| We use PCR primers elucidated from mycoplasma 16s rRNA to amplify a 475 bp amplicon as positive indication of mycoplasma presence. The primers were validated by comparing the results from thousands of cell samples tested by PCR and Hoechst 33258 mycoplasma staining. Complete correspondence were observed between those two methods to cover mycoplasma strains commonly seen in cell cultures including: M. hyorhinis, M. orale, M. arginini, M. fermentans, M. hominis, A. laidlawii and M. pneumoniae A positive DNA template amplified with amplicon of 700 b is added as method control for each sample to ensure no false negative readout. | Sample Handling |
| • Culture medium - Cultures should be grown at normal condition for at least 48 hours with cell confluency > 80% at the time of harvest. Transfer 100 ul of medium to 0.5 ml tube (screw cap tube is prefered. Regular snap cap tube should be sealed with parafilm.) There is no need to withdraw antibiotics. Samples can be shipped to us at room temperature. The sensitivity cam reach 1 - 10 copies/ul for samples in medium form. |
| • Suspension cells - can be transferred directly into 0.5 ml tubes. Spin down cell briefly and remove medium completely. The cell pellet with at least 1000 cells can be shipped to us at room temperature. |
| • Attached cells - should be trypsinized down. Neutralize trypsin by serum containing medium. Spin down cell briefly and remove medium completely. The cell pellet with at least 1000 cells can be shipped to us at room temperature. |
| • Frozen cells - should be thawn at 42°C quickly. Spin down cell briefly and remove medium completely. The cell pellet with at least 1000 cells can be shipped to us at room temperature. |
| • As few as 100 mycoplasma copies can be detected with each cell samples. The sensitivity tested on cell samples is improved compare to medium samples due to concentration. But practically, the medium sample acquired as instructed is more than adequate and more convenience. |
